Postdoctoral Researcher to perform structural analysis of Gamma secretase complexes

VIB

Leuven
Il y a 15 jours
Leuven
Il y a 15 jours

Job description

A postdoctoral position is available in the laboratory of Prof Lucía Chávez-Gutiérrez (VIB-KU Leuven) to investigate the structural strategies and molecular mechanisms operating in the γ-secretases intramembrane proteases (GSECs) in normal and disease conditions.

GSECs are uniquely distinguished by a multimeric, non-covalent structure and a sequential proteolytic cleavage mechanism. GSECs are involved in a wide number of physiological processes and also intimately connected to disease. More than 200 missense mutations in these protease complexes are linked to early onset Alzheimer’s disease (AD). In addition, dysregulation of GSECs occurs in cancer.

GSEC are thus unique multifaceted proteolytic system, crossroads for numerous pathophysiological processes. Nevertheless, central questions related to GSEC proteolytic mechanisms remain unaddressed.

This project aims at generating relevant structural and functional insights into GSEC biology.

It involves protein expression of membrane and soluble proteins using diverse recombinant systems and methodologies, protein biochemistry and substantial cryo-electron microscopy (cryo-EM). The project will be developed in close collaboration with Prof. Rouslan Efremov (VIB-VUB).

The VIB offers a vibrant, international scientific environment with access to state-of-the- art facilities; highly relevant for this project is the cryoEM facility: https://vib.be/labs/cryo-em-facility/research.

  • You have a good understanding of modern structural biology methods and experience in cryo-EM
  • You are fluent in English (verbally and in writing)
  • You like to innovative
  • You are able to work independently and as part of a team

Desirable but not required

  • Experience in protein purification of membrane proteins and their reconstitution in native-like conditions
  • Experience in standard cell culture and molecular biology techniques (cloning, protein expression, western blotting, ELISA, …)
  • Experience in protein engineering

We offer

  • An stimulating, diverse and international research environment
  • Access to state-of-the-art infrastructure

Postdoc position: Microglia-synapse molecular interactions in neurodegenerative disorders

VIB

Antwerp
Il y a 21 jours

Il y a 21 jours

About the labs

The Mancuso lab (Microglia and Inflammation in Neurological Disorders, MIND) at the VIB-UAntwerp Center for Molecular Neurology and the De Wit lab (Laboratory of Synapse Biology) at the VIB-KU Leuven Center for Brain & Disease Research, Belgium, are seeking a highly motivated postdoctoral fellow to work on a project on microglia-synapse interactions in neurodegenerative disorders.

About the project

Microglia and neuroinflammation play a central role in the pathogenesis of Alzheimer’s disease and other neurodegenerative disorders. Microglia interactions with synapses are essential for neural circuit development and function, but the role and molecular mechanisms of microglia-synapse interactions in the pathogenesis of neurodegenerative disorders remain poorly understood. This project will use mouse models, multi-omics approaches, iPSC, and cutting-edge humanized mouse systems to unravel how microglia-synapse interactions contribute to the initiation and perpetuation of neurodegenerative disease.

Your job

This is a project at the interface of two labs: the Mancuso lab, working on human and mouse microglia biology in brain disorders, and the Laboratory of Synapse Biology working on the molecular mechanisms that control precise synaptic connectivity in healthy and diseased neural circuits (see references below).

In this project, you will make use of innovative multi-omics approaches, genome engineering, viral vector technology, iPSC, and humanized mouse technology to unravel the role and molecular mechanisms of microglia-synapse interactions in neurodegenerative disorders.

The successful applicant will work in both labs and divide the time between the two sites.
